FLUOROPROFILETM PROTEIN QUANTIFICATION KITFluoroProfile is designed for the accurate and sensitive determination of proteins. FluoroProfile is based on Fluorotechnics proprietary fluorophore epicocconone which fluoresces intensely red on binding to proteins (Coghlan et al, 2005). Binding is reversible rendering stained protein completely amenable to mass spectrometry, N-term sequencing and to functional assays. FluoroProfile is robust to substances that inhibit most protein assays and is particularly suited to quantifying proteins in samples prepared for 2-D and 1-D electrophoresis, IEF gels and anywhere accurate quantification of protein is needed.

Fig 1. Assay example. BSA was prepared in water at a concentration of 1.64 mg/mL and diluted four-fold to 40 ng/mL. Aliquots (50 µL) were added, in triplicate, to a 96 well plate and 50-µL aliquots of the working solution of FluoroProfile were added. The plate was left for 40 min at room temperature and then imaged on a Molecular Dynamics Typhoon. a) an image from the Typhoon scanner of the data-point cells used to produce Figure 1.b). The image is similar to that seen when the plate is placed over a UV transilluminator. b) a plot of the fluorescence volumes against protein concentration and the corresponding correlation coefficient. FluoroProfile is equally suited to use in Xenon-arc based imagers such the BMG Fluostar.
